I guess you want to use the MKIII internal DVK, right ?

Check you enabled the integrated DVK in the MK/MKII/MKIII... properties 
dialog (in Options menu).

And check you also set up a (virtual) COM port to the MKIII protocol in 
the interfaces dialog (in Options menu as well).
